as “radiomimetic― because, like X-rays, these agents give rise to mitotic death (7) ; that is, dividing cells are “steri lized―in the sense that they are prevented from undergoing indefinite duplication, and an individual cell loses the ca pacity to form a clone. After sustaining a lethal injury, defined as an injury causing inability to form a clone, cells may divide several times and can persist and metabolize for long periods, during which they are recognized as living by physiologic tests such as dye uptake. To cause out right death, called interphase death (1 , 7), very much larger doses are required ; the mechanism involved is en tirely different, and the relative effectiveness of different substances is not the same for the 2 types of death. When drugs are used to treat tumors in vivo, the dose levels at tamable are such as to kill by the mitotic pathway (1, 7).

On the mechanism of action of the alkylating agents. II. Effects of nitrogen mustard, Myleran, and x-radiation on nucleic acid biosynthesis.

The effects of nitrogen mustard, Myleran, and x-radiation on the incorporation of isotopic orotic acid, aspartic acid, formaldehyde, adenine, and phosphate into the nucleic acids of the L1210 ascites leukemia are reported. The lack of uniformity in the incorporation pattern following pretreatment with each of the three drugs was considered significant. The treatments could result in stimulation...

Assessment of Cytotoxicity of Dimethyl Sulfoxide in Human Hematopoietic Tumor Cell Lines

Background: Dimethyl Sulfoxide (DMSO) is a solvent most broadly used as a cryopreservative agent. Antitumor effects of DMSO is a recently recognized phenomenon. In this study, cytotoxic effects of DMSO on human monocytes and T leukemic cell lines has been investigated in vitro. Methods: Human leukemic T cells (Molt-4 and Jurkat) and monocytes (U937 and THP1) were cultured in complete RPMI medi...

Chemicals and X-Rays in Mice1

Young adult female RF mice were subjected to four successive midlethal doses of nitrogen mustard (HN2), triethylene melamine (TEM), 1 ,4-dimethanesulfonoxy butane (myleran), or whole-body x-rays. The agents were given at 14-day intervals, and the survivors were observed for effects on longevity and on the incidence of late occurring diseases. The percentages of mice dying within 30 days after t...

Gnidilatimonoein from Daphne mucronata induced differentiation and apoptosis in leukemia cell lines

Among various treatments, plants play a crucial role in cancer chemotherapy. Base on literature data, different species of the thymeleaeceae family have been found to be good sources for anticancer agents. Therefore, our laboratory has initiated a research project on evaluating the anticancer properties of the Iranian medical plant, Daphne mucronata (Thymeleaeceae). Gnidilatimonoein is a new di...
